シスプラチンは、多くのがん治療で使用される代表的な抗がん薬ですが、副作用として骨格筋量や筋力の低下(筋萎縮)を引き起こすことが問題となっています。本研究では、シスプラチンによる筋萎縮の発症機序に着目し、小胞体ストレスとタンパク質合成制御との関連を解析しました。その結果、小胞体ストレスにより誘導されるDdit4/REDD1がmTORC1シグナルを抑制し、タンパク質合成を低下させることで筋萎縮が進行することを明らかにしました。また、小胞体ストレスを抑制する化合物TUDCAは、筋量および筋力の低下を有意に改善しました。本研究成果は、シスプラチン誘発性骨格筋萎縮の分子機序の理解を深めるとともに、抗がん薬治療に伴う筋萎縮の予防・治療法の開発につながることが期待されます。
Cisplatin is a widely used anticancer drug for the treatment of various cancers; however, it can cause skeletal muscle loss and weakness (muscle atrophy) as adverse effects. In this study, we investigated the mechanisms underlying cisplatin-induced muscle atrophy, focusing on the relationship between endoplasmic reticulum (ER) stress and the regulation of protein synthesis. We found that ER stress-induced Ddit4/REDD1 suppresses mTORC1 signaling, leading to reduced protein synthesis and the progression of muscle atrophy. Furthermore, TUDCA, a compound that alleviates ER stress, significantly attenuated cisplatin-induced reductions in muscle mass and muscle strength. These findings provide new insights into the molecular mechanisms underlying cisplatin-induced skeletal muscle atrophy and may contribute to the development of preventive and therapeutic strategies for muscle atrophy associated with anticancer treatment.
